Analytical Method Development and Validation of Eleutherosides Using HPTLC Technique

Abstract


For both qualitative and quantitative investigation of pharmaceutical and herbal chemicals, High-Performance Thin-Layer Chromatography (HPTLC) is a flexible and trustworthy analytical method. The invention and validation of a straightforward, reliable, and accurate HPTLC method for the measurement of eleutherosides in herbal formulations is the main goal of the current study. Ether, ethyl acetate, and ortho-phosphoric acid (6:3.5:0.5 v/v) were used as the mobile phase and silica gel 60 F254 aluminum plates as the stationary phase to accomplish chromatographic separation. At 220 nm, densitometric analysis was performed.

With a correlation coefficient of 0.9991, the method showed good breadth in the concentration range of 500–3000 ng/spot. In accordance with ICH recommendations, validation criteria such as accuracy, precision, repeatability, robustness, and sensitivity were assessed. The results showed that the limits of quantitation (LOQ) and detection (LOD) were 706.14 ng and 233.02 ng, respectively. Eleutherosides in herbal formulations were effectively analyzed using the devised approach, which demonstrated its dependability, sensitivity, and suitability for regular quality control analysis.
